If it's a daily league, then yes, but if it's a weekly league (like this one: fleaflicker.com) then no, you only set the roster once, typically early in the day on Mondays before any players have their first games of the week (at which time they lock). After players lock with the first game, they will stay in place in the starting lineup on the bench, and only commissioners can move them if needed using League Settings > Set Starting Lineups.
